Culture medium supplies nutrients and helps control the environment around cells. Its ingredients, preparation, and storage can affect growth and treatment responses. This chapter explains major medium components and compares basal, complete, serum-free, and chemically defined formulations. It connects pH, gases, temperature, and osmolality to cell behavior. Worked calculations show how final volume and dilution affect the finished mixture.

    Learning Objectives
    • After completing this chapter, you should be able to:

      1. Explain the roles of major nutrients, salts, buffers, and supplements in culture medium.

      2. Distinguish basal, complete, serum-free, and chemically defined media.

      3. Connect pH, carbon dioxide, temperature, and osmolality with cell behavior.

      4. Select medium using information about the cell source and intended experiment.

      5. Calculate stock additions, final concentrations, and simple serial dilutions.

      6. Evaluate medium preparation, storage, and culture observations using appropriate records and checks.
