Starting a culture requires more than placing living cells into fresh medium. Cells must adapt, survive, and show the behavior needed for their intended use. This chapter compares establishing cultures from tissue with recovering cells from frozen storage. It explains how dissociation, thawing, cryoprotectant handling, and starting density affect the resulting population. Viability, attachment, growth, identity, and contamination checks provide complementary evidence for deciding whether a new culture is ready for further work.

    Learning Objectives
    • After completing this chapter, you should be able to:

      1. Compare establishing a primary culture from tissue with recovering a stored cell population.

      2. Explain how tissue dissociation, medium, and surface conditions influence the cells that grow.

      3. Describe the purpose of prompt thawing and cell-specific cryoprotectant handling.

      4. Calculate starting cell concentration or seeding density from measured viable cell numbers.

      5. Evaluate recovery using viability, appearance, attachment when applicable, and growth over time.

      6. Create a traceable culture establishment record and identify evidence needed before experimental use.
