    Formative Questions    

    1. How is plasmid DNA different from the bacterial chromosome?
    2. What are competent bacteria? How can competency be artificially generated if necessary?
    3. How does pAMP allow bacteria to survive in the presence of ampicillin? 
    4. What are restriction enzymes? Restriction sites?

    Summative Questions    

    1. Define bacterial transformation.
    2. On which of the four plates will only the antibiotic-resistant bacteria grow? Explain.
    3. Calculate the transformation efficiency (TE; expressed as the number of antibiotic-resistant colonies per microgram of pAMP) for your bacteria. How does your TE compare with the average for our class? 
    4. Describe the steps required to create a recombinant plasmid.
