In this lab, students will be introduced to microscopy and related procedures, including light microscopes, electron microscopes, staining, dark-field microscopy, and phase-contrast microscopy. Students will learn to identify the purpose of each microscopy type, measure organisms using the ocular micrometer, and demonstrate their ability to view organisms under oil immersion and proper use of the phase-contrast settings.
Tasks
Students will complete the following exercises during this lab:
- View a prepared slide of Amoeba proteus using light microscopy and use the ocular micrometer to measure the organism under 40x, 100x, and 400x total magnification.
- View and measure a bacterium under the oil immersion lens.
- View live specimens using phase-contrast microscopy.
- Complete the post-lab report and submit to the instructor before leaving the lab.
The instructor should conduct a brief review of the proper use of the microscope and the types of microscopy before the lab begins. Remind students not to use the coarse adjustment knob when viewing speciments under 400x and 1000x total magnfication.
Students will also need an introduction to the purpose and proper technique related to the use of the oil immersion lens and the ocular micrometer. The position of the micrometer is usually fixed on the microscope and found under the right ocular lens; the pointer should be found in the left ocular lens. Remind students the differences between lens paper and kimwipes. Students will need to use lens paper to remove oil from the 100x (and usually 40x as well) objective lenses. Students will also need a review of the phase-contrast settings for the microscopes in our laboratory. The proper settings are listed on each of the objective lenses--students simply match the setting listed on the objective lens with the phase-contrast setting found on the condenser turret.
Although most experiments in this course are conducted in groups. This laboratory should be completed individually in order to allow students to maximize their time working with the microscope. Encourage students to discuss potential answers to each of the lab questions with one another.
