    Materials

    2 Groups per Table:

    • Organisms used from the Student Stock Cultures(TSB):
      • Staphylococcus saprophyticus
      • Escherichia coli
      • Alcaligenes viscolactis
      • Lactococcus lactis
    • 4 tubes: TSB pH 4.0
    • 4 tubes: TSB pH 6.0
    • 4 tubes: TSB pH 8.0
    • 4 tubes: TSB pH 10.0

    Procedures

    Each Group:

    1. Use a sterile transfer pipette to inoculate each of the 4 organisms into each pH tube:
      • add 1 drop of broth culture to each pH broth tube.
      • Staphylococcus saprophyticus → TSB pH 4.0, pH 6.0, pH 8.0, pH 10.0
      • Escherichia coli → TSB pH 4.0, pH 6.0, pH 8.0, pH 10.0
      • Alcaligenes viscolactis → TSB pH 4.0, pH 6.0, pH 8.0, pH 10.0
      • Lactococcus lactis → TSB pH 4.0, pH 6.0, pH 8.0, pH 10.0
    2. Place the tubes in buckets and incubate in your classroom incubator.

    Results

    Enter your ratings for each bacterium/pH and give the pH classification for bacterium.
